The NucleoCounter® NC-250™ is an automated cell analyzer utilizing fluorescence imaging to characterize cell properties. The NC-250™ can perform high speed cell count and viability determination, a unique 5-minute cell cycle assay and a 1 minute apoptosis assay. The accompanying intuitive software can be used for data analysis of the results and has unlimited licenses. The NC-250™ benchtop image cytometer ensures high precision while being calibration free.

Cell Counting assays
With the combination of simplicity and innovation in the NucleoCounter® NC-250™, counting and viability determination of mammalian cells have never been more uncomplicated. Simply mix a small volume of cell suspension with fluorescent dyes, load and run. Within 3 minutes you will have 8 high precision viability and cell count determinations.
With the NC-250™, 8 viability and cell counts can be performed in less than 3 minutes. Only 10 µL of a mixture of cell suspension and fluorescent dyes is loaded to the chambers in A8 slides. For cells with low concentration or for even higher precision, the A2 slides with 30 µL loaded cell mixture can be used.
With the NucleoCounter® NC-250™, ChemoMetec A/S offers a wide range of optimized protocols meeting the needs of all mammalian cell types grown under different conditions. Through unique assays cell count can be performed even on cells growing on microcarriers and in sphere cultures.

Even for simple cell count and viability assays, it is essential to evaluate the obtained result. With the intuitive NucleoView™ software for NucleoCounter® NC-250™, it is possible to define the counted events precisely. The linkage between images and plots showing fluorescence intensity allows for a precise definition of the events to count – for both total and dead cell populations.
The accompanying NucleoView™ software allows for easy coupling between the obtained image and plots showing fluorescence for precise determination of the events to count.

The vitality assay in NucleoCounter® NC-250™ is an extremely fast and simple assay for evaluation of cellular health, oxidative stress and detection of late-stage apoptosis. The fluorescent dye VitaBright 48™ detects reduced thiols such as e.g. glutathione. During late-stage apoptosis, the level of reduced thiols decreases resulting in a lower VitaBright 48™ intensity. With the accompanying user-friendly software for NC-250™, advanced data analysis can effortlessly be performed. The unique possibility to link between images, histograms and scatterplots allows for detailed and precise data analysis.
Example of data analysis of YAC-1 cells grown in the absence (upper row) or presence of camptothecin, CPT (lower row). Cells were stained with VB-48™ and Propidium Iodide (PI) and analyzed using the vitality assay and a NucleoCounter® NC-250™. Scatter plots and histograms were obtained from the NucleoView™ NC-250™ software.
