The NucleoCounter® SCC-100™ Somatic Cell Counter is a high-quality image cytometer capable of measuring the number of somatic cells in a milk sample with high precision and accuracy. The instrument itself is extremely simple to operate, even for the untrained operator. When measured, the simple media is contained in the SCC-Cassette™, an innovative disposable device. Add reagent C to the sample, load the cassette and press run and the result is presented within 30 seconds.
The measurement range of the NucleoCounter® SCC-100™ is between 1×104 Cells/ml to 2×106 Cells/ml., with an optimal measurement range of between 1×104 Cells/ml to 1×106 Cells/ml.
The NucleoCounter® SCC-100™ can be connected to a PC using USB port. It must be noted that all calculations, cell counting and data analysis are performed in the NucleoCounter® SCC-100™ which is a true stand-alone instrument. The transfer of image data to a PC provides the user with additional documentation options.

NucleoCounter® SCC-100™ – Highest Precision in Counting Somatic Cells
Reproducibility of the NucleoCounter® SCC-100™
The inherent properties of the ChemoMetec Image Cytometry implementation offer unique cell counting properties, the most important one being outstanding reproducibility performance. Each instrument is calibrated during production and the durable properties of the LED light source, optical system and the image capturing camera means that no additional calibration is required, neither short-time nor long-time calibration.
This is demonstrated in the histogram below which shows the results of the comparison between 51 NucleoCounter® SCC-100™ instruments and a NucleoCounter® SCC-100™ reference instrument. The reproducibility was tested by measuring a number of milk samples on each of the instruments being tested as well as on the reference instrument. The histogram illustrates the observed relative difference in Cell Count, where the observed mean difference is -0.011% and the standard deviation of differences of 0.46%, which is comparable with the expected standard deviation of the mean, calculated to be 0.65%.
Comparison between the NucleoCounter® SCC-100™ and a Flow Cytometer
Raw cow milk samples were measured on NucleoCounter® SCC-100™ and “Industry Standard” Flow Cytometer. Samples were heated to 40°C and measured on the Flow Cytometer. 500 µL of a milk sample at room temperature was mixed with 500 µL of Reagent C and measured on the NucleoCounter® SCC-100™. The figure below demonstrates the excellent correlation between the two methods in the range up to 1,000,000 Cells/mL.
