How to Create Extra Tables and Charts in Plot Manager for the NC-200, NC-250, and NC-3000

Background

For some assays on the NucleoCounter series such as Flexicyte on the NC-3000 it is necessary to add tables and charts to interpret your data. For others you may wish to add a custom table or chart to aid in the analysis of your data.

How To Create Extra Tables and Charts in Plot Manager for the NC-200, NC-250, and NC-3000

  • Open the Plot Manager by selecting Tools > Plot Manager
  •  
  • Open the File Browser by clicking the small folder icon to the right of the Data Folders and Files View
  • Select any data files you wish to add a chart to by highlighting and dragging them into the Plot Manager
  • On the data file you wish to add to, please select whether you wish to add a Table, Histogram, or Scatterplot and then please find the appropriate section below

Creating Tables in Plot Manager for the NC-200, NC-250, and NC-3000

  • On the data row you wish to add a Table to please click the “Add a New Table Plot” Button on the far left hand side of the row
  • Once the new blank Table has been added to the row, double click it to enlarge the area, and then select “New Table” at the top
  • Selecting the “New Table” Option will allow you to set the upper left corner of your table, as well as the number of rows and columns present.
  • Once your table location and size is set, you may click in the blank boxes to begin inputting information
  • To pull numerical information from the file you are adding the Table to, click the blank box and start by inputting an “=” sign, this will open the option to insert value.
  • This information will be anything collected during the protocol, such as number of events in a given gate, or volume measured. Using these you may make your own formulas to analyze data as you see fit.

Creating Histograms in Plot Manager for the NC-200, NC-250, and NC-3000

  • On the data row you wish to add a Histogram to please click the “Add a New Histogram Plot” Button on the far left hand side of the row
  • After clicking the “Add a New Histogram Plot” button, a histogram will populate in the row selected with default axis settings
  • To change the axes, please click on the axis label you wish to change and a menu of options will appear, allowing you to set the plot to your data set of choice
  • You may also set a marker to determine the proportion of data between two points. This is performed by selecting the “New Marker” option at the top of the plot window followed by clicking where you would like the outer bounds of the marker to be

Creating Scatter Plots in Plot Manager for the NC-200, NC-250, and NC-3000

  • On the data row you wish to add a Scatter Plot to please click the “Add New Scatter Plot” Button on the far left hand side of the row
  • After selecting “Add New Scatter Plot” a scatter plot will populate in the row selected with default axis settings.

To change the axes, please click on the axis label you wish to change and a menu of options will appear, allowing you to set the plot to your data set of choice

  • Once your Axes are set up how you would like them, you may establish gating in two ways to help quantify your data through the options at the top of the screen “New Polygon” or “New Quadrent”
  • For “New Polygon” you will click the vertices of a shape drawn around your data as seen below, ensuring that the final point you click is the starting point, so the shape is closed.
  • This shape can be changed by selecting the completed polygon, turning it red, and then dragging the vertices to the location you would like them to be
  • To add a quadrant, please select the “New Quadrant” option at the top of the window and then click where you wish the center of the quadrants to be as shown below.

The boundaries for the quadrants can be changed similarly to a polygon, by clicking the quadrant boundary once it will turn red and can be drug to the position you wish it to be at.
