How to export NC-3000 Data to FCS and ACS files

Background

FCS is a data file standard used for writing and reading flow cytometry experiment data; contains one or more data sets, which are a collection of information produced by a cytometer. The FSC standard is developed and maintained by the International Society for Advancement of Cytometry (ISAC).

FCS files may be used by those who wish to perform data analysis using third party analysis software such as FCS Express, De Novo Software or FlowJo, BD Bioscience. 

NOTE: Data can also be exported as ACS format which is compatible with FCS Express 4 Image Cytometry, De Novo Software.

How to export data to FCS files

  1. Export single file
    1. Open NucleoView NC-3000 software click on the required data file

2. Go to the File Tab

3. Click on “Export data” the following dialog box appears

4. Make sure FCS is in the “Export format” window, toggle down if it does not appear and click on it. 
5. Make sure all the parameters are checked, they are selected by default. 
6. Click Export. 
Note: a dedicated folder may be created prior to the export and selected under “Export File Name”.

7. To find your file, go to the FCS File folder in NucleoView NC-3000 folder, your selected file will now have .FCS at the end and you can also find it under Type as FCS files.

  • Export multiple files
    • Multiple files can be batch exported all at once. 

**The files must have been acquired using the same protocol.**

  • In the main window, click the file browser in the upper right corner the following dialog box opens

• Select the files to export

• Right click a dialog box appears, select “Batch Export Data”

• A dialog box appears

• Make sure FCS is in the “Export format” window, toggle down if it does not appear and click on it. 
• Make sure all the parameters are checked, they are selected by default. Click Export. 

Note: a dedicated folder may be created prior to the export and selected under “Export File Name”.

  • The files can be found in the selected folder.
  • Export ACS files
    • All files can be exported as ACS following the same steps given above. Make sure ACS is selected as “Export Format”
    • In order to view the images in FCS Express 4 Image, make sure that the Load Images option in under Edit Preferences->Data Loading->ICE format in FCS Express 4 Image is checked.
