How to set up a gate for the cell diameter

Background

The actual cell diameter that is displayed under the results tab will display the cell diameter from most events in the first AO size vs. AO intensity plot[1], independently of the standard P1 gate, meaning it is including both, cell debris and actual cells into the analysis. But it is possible to determine the exact cell diameter of a certain cell population, e.g. only the cells which are displayed in the P1 standard counting gate.


How to set up a gate and calculate the cell diameter

  • Go to the cell diameter plot and select “new marker”. Set up the marker e.g. for the cell debris. Close the plot by choosing ok.
  • Go to the table plot and generate a new table.
    • Click right in the table and select “insert formula – insert value – gate parameter – plot 3 – marker – m1 – mean-x”. Press “enter”.
    • Click right again and select “format cell”. Choose decimal places and add a symbol, e.g. µm.
  • Optional: Save your changings as a new protocol.

How to set up a gate and calculate the cell diameter only for cells in P1

  • Go to the cell diameter plot and select “gating setup”. Choose “P1 AO gate – include” and choose “apply – ok”. Now only P1 cells are displayed in the cell diameter histogram.
  • Select “new marker”. Set up the marker across the whole plot.
  • Go to the table plot and generate a new table.
    • Click right in the table and select “insert formula – insert value – gate parameter – plot 3 – marker – m1 – mean-x”. Press “enter”.
    • Click right again and select “format cell”. Choose decimal places and add a symbol, e.g. µm.
  • Optional: Save your changings as a new protocol.
